Javona Glover, 23, was killed outside a Walgreens in Tallahassee, local police confirmed to The Associated Press.
The singer's niece has a 2-year-old child, and T-Pain took to Instagram to post a tribute photo montage of her with her baby.
Fans responded with messages of condolences.
"Terrible tragedy" and "stay strong" were just a couple of the sentiments shared by fans on his page.
After posting early Tuesday, requesting any information on the homicide, T-Pain, 30, took to Twitter to thank fans and friends for support.
According to The AP, police have identified a suspect and are still investigating the crime.
